O Before the unit is started up for the first time, it is important to make sure that the voltage and frequency indicated on the label of the unit correspond to those on the mains! O Avant la premiere mise en marche, il est important de verifier que le voltage et la frequence indiques sur le label de l'appareil correspondent au reseau electrique. O If dangerous substances are to be pumped off during a freeze-drying process, suitable and appropriate safety measures must be taken and the corresponding safety rules followed (e.g. pumping off into an exhaust air system). O To be able to ensure safe operation, only original FINN-AQUA components must be used. O For service purposes, the unit may be opened only by trained personnel. Do not forget to pull the plug from the mains before beginning with any manipulation of the unit! In keeping with EG guideline 360, 1976/79 and VBG 16, for reasons of safety and for the protection of the environment, it is important to indicate or declare any dangerous substance which may be around or inside the unit. Without a declaration in this respect, FINN-AQUA will not carry out any service work. We reserve the right to make changes in the unit without prior notice, if such changes lead to improvements in the design and performance of the unit. Description of the Basic Unit 2.Description de l'appareil de base The LYOVAC GT 2 is a table-top freeze-dryer. The vacuum pump (16) can stand either on the floor or on the bottom shelf of the trolley (52). It is connected to the basicunit (1) by means of a vacuum line (18). This freeze-dryer is very versatile because of its modular design. Using the acrylic glass cylinders - 185 (2) and 440 (10) - it is possible lo operate following the one-chamber system for drying with the manifold (Fig. 2), or following the two-chamber principle for drying both with the manifold and with trays (Fig. 1 and Fig. 4). The possibilities offered by the numerous available accessories (e.g., stoppering device, spin-freezing, cooling and heating of shelves) are described in detail in Point 9, page 34). L'appareil LYOVAC GT2 est prevu pour etre dispose sur une table de laboratoire. La pompe a vide (16) est placee a cote de l'appareil de base ou eventuellement sur une etagere sous l'appareil lorsque celui-ci est dispose sur un chariot (52) adequat. La pompe a vide est reliee a l'appareil de base (1) au moyen du flexible (18). La conception modulaire de cet appareil permet de nombreuses combinaisons d'application. The unit is evacuated by means of a two-stage vacuum pump Type TRIVAC D 4 B or D 8 B (16) which has its gas ballast valve open during the evacuation process. As soon as the pump is switched off, the suction port valve fitted in the vacuum pump (16) closes automatically and the pump is vented at the same time. The pressure in the chamber rises either because of a leak in the system and/or because of the gas being released slowly by the product. (See operating instructions of the vacuum pump). The condenser coils (19) are cooled by means of a onestage cooling compressor (20). The condensing surface is designed in such a way that the large amounts of water va•' por which arise at the beginning of the process can be condensed. It is possible to dry certain products on trays (9) directly between the coils of the condenser tube (19) (see Point 7.1.2, page 21). When the unit (1) is switched off, the condenser coils (19) are isolated from the vacuum pump (16) by the suction port valve. After venting, the ice on the condenser coils (19) warms up and slowly begins to melt. More rapid defrosting can be attained by pouring water over the condenser coils. During the defrosting process, the basic components of the unit (base plate (3), chamber (2) (10), etc.) are put at the back of the basic unit (1). The cooling unit, consisting of a one-stage, air-cooled cooling compressor (20), operates with refrigerant R 502 and is used to cool the condenser coils (19). When the FREEZING button (50) is pressed, the compressor (20) is switched on after a time delay. If the DRYING button (51) is pressed, the vacuum pump (16) is also switched on. Pressure measurements are carried out by means of a vacuum gauge with a measuring range from ambient pressure to 10"3 mbar. The gauge is integrated into the operating panel (22) of the basic unit (1). (See Fig. 5). En ce qui concerne les autres possibilites de montage d'accessoires (par ex. syteme de fermeture, systeme de rotation ou plateaux de chargement thermostatises) que nous pouvons vous offrir; ceux-ci seront developpes au pt. 9, p. 34. Cet appareil est mis sous vide au moyen d'une pompe a vide bi-etagee avec lest d'air ouvert du type TRI VAC D4B/D8B (16). Ce type de pompe a vide possede une vanne de securite qui se ferme automatiquement en cas d'arret de la pompe a vide. La pression regnant dans la chambre de dessiccation depend done de l'etancheite de l'appareil et/ou de la desorption lente du produit. La pompe a vide est remise a l'air automatiquement hors tension, (voir mode d'emploi de la pompe a vide). Le condenseur (19) est refroidi par un groupe frigorifique a un etage. La surface de condensation a ete etablie afin d'obtenir la plus grande quantite de vapeur d'eau des le debut de la sublimation. Les produits a lyophiliser peuvent etre egalement congeles sur des plateaux disposes directement entre les serpentins du condenseur. (voir pt. 7.1.2., p. 21). Lorsqu'on arrete l'appareil, le condenseur se trouve isole grace a la vanne de securite integree a 1'aspiration de la pompe a vide. Lors de la remise a Fair de l'appareil, la glace disposee sur le condenseur se rechauffe et fond lentement. Un degivrage rapide peut etre realise a l'eau. Le groupe frigorifique a un etage, refroidi a l'air (20) est alimente avec du freon R 502 et il refroidit le condenseur. En phase ,,FREEZING" le groupe frigorifique se met en marche. Lorsqu'on appuie sur la touche ,,DRYING", la pompe a vide (16) se met en marche. La mesure de la pression est obtenue au moyen d'un appareil de mesure de vide integre dans le module de commande (22) de l'appareil de base et peut atteindre des pressions jusqu' a 10° mbar (voir fig. 5). 3.Preparing for Start-Up (First Operation) 3.Preparation pour une premiere mise en marche 3.1 Checking the Scope of Delivery 3.1. The parts must be taken out and prepared for assembly in the same sequence in which they are mentioned in the operating instructions (see, too, Point 9 - Description of Accessories). "®" For this reason, check the catalogue numbers of the parts delivered in the same sequence as they appear in the operating manual. 3.2 Assembly of Basic Unit (1), Vacuum Pump (16) and Exhaust Filter (17) ..3.2.1 Setting Up the LYOVAC GT 2 Basic Unit (1) 3.2. Avoid direct sunlight on the condenser coils (19) or too high an ambient temperature either of which could impair the refrigerating capacity considerably. Installation de la pompe a vide TRIVAC D 4 B ouD,8B 3.2.2 Setting Up the TRIVAC D 4 B or D 8 B Vacuum Pump (16) • Deballer la pompe a vide (16) TRIVAC D4B (cat n° 112 45) ou TRIVAC D 8 B (cat n° 112 55). / Unpack the TRIVAC D 4 B (Cat. No. 112 45) or the TRIVAC D 8 B (Cat. No. 112 55) vacuum pump (16). Remarque: d'autres pompes a vide peuvent etre adaptee sur notre appareil a condition que les donnees technique soient identiques. Note: Be sure to follow the instructions given in the operating manual of the vacuum pump. 3.2.3 Connecting an Exhaust Filter Type AF 4-8 (17) or an Exhaust Filter with Oil Return Type AR4-8 / Unpack the AF 4-8 (Cat. No. 189 06) exhaust filter (17) or the AR 4-8 (Cat. No. 189 20). / Check the scope of delivery of the exhaust filter (17) which includes the following: - 1 AF4-8orAR4-8(17) - 1 Operating manual for the AF 4-8 or AR 4-8. - 1 TRIVAC D 4 B/D 8 B avec cordon de raccordement electrique - 1 mode d'emploi TRIVAC D 4 B/D 8 B / Installer la pompe a vide (soit a cote de l'appareil, soit sur le chariot de laboratoire) Attention: Bien lire le mode d'emploi de la pompe a vide avant de l'utiliser. 3.2.3. Monter le filtre d'echappement AF 4/8 ou le filtre d'echappement avec retour d'huile AR 4-8 / Deballer le filtre d'echappement (17) AF 4/8 (cat n° 189 06) ou AR 4-8 (cat n° 189 20) / Contrule de livraison du filtre d'echappement - 1 AF 4-8 ou AR 4-8 - 1 mode d'emploi AF 4/8 ou AR 4/8 / Monter le filtre d'echappement a la bride d'echappement de la pompe a vide 3 Vorbereitung zur ersten Inbetriebnahme / Auspuff-Filter (17) an Druckseite der Vakuumpumpe (16) anschlieBen. Anmerkung: Gebrauchsanleitung (GA) des Auspuff-Filters beachten! 3.2.4 Elektrische und vakuumtechnische Verbindung zwischen Grundanlage (1), Vakuumpumpe (16) und Auspuff-Filter (17) ACHTUNG! Alle Dichtflachen vor der Montage auf Sauberkeit priifen! Abb. 6 Riickseite Grundgerat / Vakuumleitung (18) der Grundanlage am Saugstutzen der Vakuumpumpe (16) montieren. Dazu a) Zentrierring DN 16 KF (Kat.-Nr. 183 26) in den Saugstutzen einlegen b) Schlauchstutzen aufsetzen c) Spannring DN 10/16 (Kat.-Nr. 183 41) urn die Verbindung legen und handiest anziehen •/ Elektnsches Netzkabel der Vakuumpumpe in die dafiir an der Riickseite der Grundanlage vorgesehene Steckdose (34) (Abb. 6) stecken. 39 ACHTUNG! Stellen Sie jetzt noch nicht die elektrische Verbindung zwischen Grundanlage (1) und elektrischem Netz her! / Wenn kein Auspuff-Filter (17) verwendet wird, sollte ein Abluftschlauch auf den Druckstutzen der Pumpe gesetzt werden. Dazu aus dem Zubehor der Vakuumpumpe a) den Zentrierring DN 16 KF (Kat.-Nr. 183 26) in den Druckstutzen einlegen b) einen Kleinflansch mit Schlauchttille DN 16 KF (Kat.Nr. 183 19) aufsetzen und c) den Spannring DN 10/16 (Kat.-Nr. 183 41) urn die Verbindung legen und handiest anziehen. Anmerkung: Der vorgenannte Kleinflansch (Kat.-Nr. 183 19) gehort nicht zum Standard-Lieferumfang. Empfehlenswert ist - der AnschluR des Abluftschlauches an eine hausinterne Absauganlage mit Abluftreinigung, oder - die Heraustuhrung des Abluftschlauches auBerhalb des Gebaudes. -* Gilt nicht fur LYOVAC GT 2-E * Not valid for the LYOVAC GT 2-E * N'est pas valable pour le LYOVAC GT 2-E Diesc Empfehlung gill auch bei Verwendung eines Auspuff-Filters (17). 3.Preparing for Start-Up (First Operation) 3.Preparation pour une premiere mise en marche / Connect the exhaust filter (17) to the pressure side of the vacuum pump (16). (BIEN VERIFIER la position d'echappement de la pompe dessinee sur la face avant de la pompe) Note: Be sure to follow the instructions given in the operating manual of the exhaust filter. 3.2.4 Effecting the Electric and Vacuum Connections between the Basic Unit (1), the Vacuum Pump (16) and the Exhaust Filter (17) Note: Make sure all the sealing surfaces are perfectly clean before beginning with assembly. / Connect the vacuum line (18) of the basic unit (1) to the suction port of the vacuum pump (16). For this, proceed as follows: a) Place the DN 16 KF centering ring (Cat. No. 183 26) in the suction port of the pump. b) Mount the hose nipples. c) Place a tension ring DN 10/16 KF (Cat. No. 183 41) around the connection and tighten firmly by hand. / Plug in the electric cable of the vacuum pump (16) into the socket (34) provided for this purpose at the rear of the basic unit (1) (Fig. 6). CAUTION Do not connect the basic unit (1) to the electricity mains yet. / If an exhaust filter (17) is not being used, an exhaust hose should be led out from the pressure port of the pump. For this, proceed as follows: a) Place the DN 16 KF centering ring (Cat. No. 183 26) in the pressure port. b) Insert a small flange with hose nipple DN 16 KF (Cat. No. 183 19). c) Place the DN 10/16 KF tension ring around the connection and tighten firmly by hand. Note: The small flange (Cat. No. 183 19) mentioned above does not form part of the standard scope of delivery. It is advisable to connect the exhaust hose to a factory exhaust line provided with an exhaust air purification system or to lead the exhaust line outside the building. These recommendations should also be followed if an exhaust filter (17) is being used. Attention: Bien lire le mode d'emploi des filtres d'echappement 3.2.4. Raccordements electriques et vide entre appareil de base, pompe a vide et filtre d'echappement Attention: - Controler que tous les joints soient propres avant le montage. </ Monter le flexible (18) de 1' appareil de base a la bride d'aspiration de la pompe a vide (voir position d'aspiration sur la face avant de la pompe a vide). Pour cela: a) placer l'anneau de centrage DN 16 KF (cat n° 183 26) sur la bride d'aspiration de la pompe a vide. b) placer le flexible sur la bride munie de l'anneau. c) fixer les deux brides au moyen du collier de serrage DN 10/16 KF (cat n° 183 41) en sen-ant la vis du collier de serrage. •/ La fiche du cordon electrique de la pompe a vide doit etre introduite dans la prise de courant ,,pompe a vide" situee a l'arriere de l'appareil de base (voir fig. 6). Attention: Ne raccordez pas encore l'appareil de base au reseau electrique. / Si vous ne disposez pas d'un filtre d'echappement pour votre pompe a vide, il est conseille de mettre un tuyau flexible a l'echappement de la pompe a vide avec les accessoires suivants: a) un anneau de centrage DN 16 KF (cat n° 183 26) a l'echappement b) une olive a ajutage crante DN 16 KF (cat n° 183 19) sur l'anneau c) fixer les deux brides par un collier de serrage DN 10/16 KF (cat n° 183 41) en vissant le papillon du collier Attention: ces accessoires ne font pas partie de la livraison standard (cat n°. 183 19) Conseil de montage : - Disposer une extremite d'un tuyau flexible sur 1'olive a ajutage crante et l'autre vers une hotte d'aspiration eventuelle du laboratoire ou vers l'exterieur du batiment. Ce montage est egalement conseille quand on dispose d'un filtre d'echappement. 7 3.Preparing for Start-Up (First Operation) 3.Preparation pour une premiere mise en marche 3.3 Mounting the Manifold for Freeze-Drying (Fig. 2) 3.3. Installation d'un appareil avec disque special pour ballons (fig. 2) Note: Make sure all the sealing surfaces are perfectly clean before beginning with assembly of the components. Attention ! Avant d'assembler tous les composants, veillez a ce que toutes les surfaces et joints soient propres. / Unpack the 440 acrylic glass cylinder (10) (Cat. No. 045 67). / Deballer le cylindre Acrylglass 440 mm (10) (cat n° 045 67) / Place an L seal (24) (Cat. CAUTION It is important to avoid having the magnet (28) attracted directly by the operating handle. The magnetic force is so great that it is then very difficult to separate the magnet again. / Connect the vacuum gauge (15) to the base plate (3). During the freeze-drying process, the vacuum pump (16) must operate with the gas ballast valve open so that the water vapor being suctioned off cannot condense in the pump thus contaminating the pump oil. The drying times depend on the layer thickness and the solids content of the product. The layer thickness, should be 15 mm or less. A uniform charging of the unit, the same layer thickness throughout and the processing of just one product at a time are strongly recommended. Le temps de sechage est dependant de l'epaisseur du produit et de la concentration du produit. L'epaisseur du produit devrait etre optimalement de 15 mm ou inferieur. The temperature down to which the product is frozen is determined by the solidification temperature of that particular product. No. 045 85) makes it possible to regulate the pressure in the chamber to maintain the desired pressure values (see Point 8, page 29).* Within limits, the pressure in the chamber can also be regulated by the heating and cooling system of the shelves.* By heating the shelves (8) (Cat. During secondary drying, the water adsorbed at the inner surfaces is pumped off at the lowest possible pressure. This makes it possible to attain a very low residual moisture in the product. Die Ventilplatte (5) (Kat.-Nr. 045 62) muB vollstandig geoffnet werden. 7.3.2 Endpunktbestimmung der Nachtrocknung mit Druckanstiegsmessung (DAM) Die Bestimmung des Trocknungsendes erfolgt, wie unter Pkt. 7.2.2, S. 23 beschrieben, durch eine Druckanstiegsmessung. 7.4 Nachbehandlung des Trockenguts Wenn der gewiinschte Trocknungsgrad erreicht ist (Restfeuchtegehalt), kann das System geflutet werden. Dieser Vorgang sollte nicht ohne besondere Vorkehrungen ablaufen. Zunachst mufi die Grundanlage (1) vom Vakuumpumpensystem getrennt werden. Dazu • das Absperrventil (32) (Kat.-Nr. 045 86) verschlieGen oder DRYING • die Taste ,,DRYING" (51) betatigen ODie Vakuumpumpe (16) und die Ka'ltemaschine (20) schalten ab. Wird das System mit Luft geflutet, kann die darin enthaltene Feuchtigkeit sofort vom Trockengut aufgenommen werden. Auch der Sauerstoffanteil kann sich bei der anschlieBenden Lagerung ungiinstig auswirken. Als MaBnahmen gegen diese unerwiinschten Wirkungen bieten sich an: - die Verwendung der Trocknungspatrone (Kat.-Nr. 042 13) am Beluftungsventil (14) - das Fluten mit trockenem Stickstoff - das VerschlieBen der Ampullen oder Flaschchen mit einer VerschluBeinrichtung (Kat.-Nr. 045 72 oder045 73) unter Vakuum oder Teildruck. Bei keimarmem Arbeiten zusatzlich: - Das Vorschalten eines Sterilfilters mit SchlauchanschluB. 7. This process, however, should not be carried out without taking preliminary precautions. Afin de vous proteger contre ces inconvenients, nous pouvons vous proposer - le placement d'un filtre special (cat n° 042 13) a placer a la vanne de remise a 1'atmosphere - de remettre a l'atmosphere au moyen d'une alimentation en azote sec. - d'obturer les ampoules ou flacons avant remise a l'at- ' mosphere au moyen de notre accessoire (cat n° 045 72 ou 045 73). Pour eviter toute pollution par germe, nous vous conseillons de disposer un filtre sterile avec embout flexible. 24 7. Dazu • die Taste ,,DRYING" (51) betatigen. ODie Vakuumpumpe (16) und die Kaltemaschine (20) schalten ab. Es ist zu empfehlen, die Vakuumpumpe (16) noch ca. 30 min nachlaufen zu lassen. Das Eis im Kondensator kann auch ohne zusatzliche Warmezufuhr abtauen. Dieses nimmt allerdings mehr Zeit in Anspruch. Dabei ist das Entfernen der Aufbauten nicht erforderlich. Das System muB beliiftet und die Ventilplatte (5) geoffnet sein, damit das Kondensat ablauft. Kontrollieren Sie vor dem Beginn der nachsten Gefriertrocknung, ob das Abtauwasser vollstandig aus dem Kondensator (19) und dem Ablaufstutzen abgeflossen ist. Anmerkung: Zwischen dem Anhangen der einzelncn Kolben sollte ein Zeitabstand von mindestens 15 Minuten bestehen, damit sich im Kolben stabile Trocknungsbedingungen einstellen konnen. In Abhangigkeit von der Anzahl der angehangten Kolben erhoht sich der Betriebsdruck. Damit ein Antauen des Produktes vermieden wird, muB er jeweils vor dem Anhangen uberpriift werden. f) Gefriertrocknung beenden An der AuBenwand der Kolben kondensiert wahrend der Trocknung Luftfeuchtigkeit. Im allgemeinen kann man davon ausgehen, daB das Produkt trocken ist, wenn O dieses Eis abgetaut ist und die Kolben Umgebungstemperatur angenommen haben. Zur Uberpriifung konnen Sie \/ mitderHandinnenflachedie KolbenauBenwandberiihren. Wenn die Trocknung beendet ist, werden • die Ventile (23) durch Drehen des Schwenkhebels um 180° (Nase nach oben zeigend) verschlossen und die Kolben beluftet. Dabei \/ die Kolben festhalten und nach unten vom Kunststoffkonus der AnschluBventile abnehmen. Vous pouvez controler en testant avec la main la t° exterieure du ballon qui devrait etre egale a la t° ambiante. The grooved stoppers necessary to stopper the vials are placed loosely on the vial necks before the drying process is begun. After drying has been completed, the pressure plate or plates are lowered manually with the cross handle. This causes the stoppers to be pressed firmly into the vial necks. Stoppering can take place under vacuum, partial vacuum or protective gas (e.g. nitrogen). Systeme de congelation en rotation This device makes it possible to freeze the product under vacuum while at the same time taking advantage of the centrifugal force. Avec cet accessoire, les produits disposes dans les ampoules sont congeles sous vide par centrifugation. The liquid product is frozen by removing the evaporation heat under vacuum. To make sure that foam or bubbles cannot form, the ampoule holder is rotated so that the centrifugal force presses the product, still in the liquid state, outward against the container walls. The product creeps up the wall and forms a large surface. Le produit liquide est congele par soutirage de la chaleur d'evaporation dans le vide. Its outer design with its aluminium profiles and shelves lacquered in black provides a well-organized work area and is, beyond that, an optically pleasant addition to the laboratory. Plug in the mains plug of the vacuum pump at the rear of the basic unit (Fig. 6, page 7). Run the pump with no load and with an open gas ballast valve until cleaning of the oil has been completed. Effect a change of oil if necessary. - The suction line has not been connected or is not properly connected. Check the small flange connection between the suction line and the vacuum pump. Has the centering ring been placed correctly? - The discharge valve (25) of the basic unit i> open. - The venting valve (14) of the base plate is open. - The chamber-condenser valve (5) is closed. Plug in the mains plug and the control line of the pressure regulating device correctly in the rear panel of the basic unit (Fig. 6, page 7). Check Fuse 3 (F 32 mA) and replace it if necessary. The RTD (31) for the temperature regulation is not in contact with the measuring lead marked SHELF, but with the line marked T2. The set temperature that has been set is too high. - The compressor (20) is not in operation or is defective. Wait until the pressure is lower than Pmax. Plug in the heating cable (54) properly to the intermediate ring (4). Check the plug connection. Check the fuses and replace them if necessary. Plug in the RTD (31) into the heatable shelf and into the leadthrough of the intermediate ring in the measuring lead (55) marked SHELF. Check the set temperature and correct it if necessary. - Plug in the RTD (31) into the retaining borehole of the heatable shelf. - Check the contacts of the RTD (31).
