1-800-477-6834 g. Reaction Ratios AA:Act:Base – Software calculates suggested amino acid to activator to base ratios. Click the button to change this value The Base section lists: a. Base Name – Click the button to change the name of the base b. Base Mole Wt – Click the button to change the molecular weight of the base (in g/mol) c. Base Density – Click the button to change the density of the base (g/mL) d. Base Total Volume (est) – Software calculates the suggested total volume of base solution (in mL) to use for the synthesis. Click the button to change this value. e. Base Weight – Software calculates amount of base (in g) to create the base solution. f. Base Volume – Software calculates the volume of base (in mL) to create the base solution. At the bottom of the screen, the Print button allows you to print the calculated values. The Return button returns you to the Sequence Editor screen. 7. Copy Fwd – Copies the programs assigned to the selected cycle (in all 4 columns) through to the last cycle. 8. Go To Pos – Select cycle position to edit 9. First – Goes to first cycle 10. Prev – Goes to previous cycle 11. Next – Goes to next cycle 12. Last – Goes to last cycle 13. Syn RVs – Select 1 or 2 14. Load RV1 – Loads current peptide settings onto RV 1 on Load Synthesis screen 15. Load RV2 – Loads current peptide settings onto RV 2 on Load Synthesis screen 16. Clear All Loaded RVs – Clears all settings on the Load Synthesis screen 17. Print – Prints the values on the Sequence Calculations screen 18. Save – Saves settings 19. Save As – Saves settings to a different file name 20. Cancel - Main Menu – Returns to Main Menu without saving changes 3.2.1 Calculate Coupling Difficulty The Sequence Editor may be used to calculate the synthesis difficulty of each coupling step for a peptide sequence. 57
